How can i plug a tire puncture from a nail AND re-inflate unmounted tire?

If it was just a nail you could buy a plug kit at a local auto part store and just fill it up but a big bolt may leave a large hole, if that is the case you need to take it to a tire store for a patch on the inside or better yet a tube.
